Chief Inspector Lorraine Dobson said: "The powder has been tested and has been described as not-hazardous.
"I would like to thank the teachers and pupils at the school for their co-operation.
"I would also like to thank our colleagues in the Fire and Ambulance Service for their assistance."
Earlier the PSNI had closed access to the school as part of an investigation at the front of the premises. The alert began at about lunchtime.
A text message from the school to parents said all students and staff were safe.
Lagan College confirmed to the Belfast Telegraph that the school day continued as normal and that the incident was "only at the front of the school".
Pupils were later allowed to leave the school supervised by staff.
